Biological Activity

Futibatinib (TAS-120) is an irreversible inhibitor of fibroblast growth factor receptors, targeting FGFR1, FGFR2 (IC50 1.4 nM), FGFR3, and FGFR4 (IC50 3.7 nM). By covalently binding to the ATP-binding pocket of FGFRs, it suppresses receptor autophosphorylation and downstream signaling pathways. Consequently, futibatinib inhibits cell proliferation and selectively induces cell death in cancer cell lines carrying FGFR genomic alterations. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
