Biological Activity

FR054 is a small-molecule inhibitor of phosphoglucomutase 3 (PGM3), a key enzyme in the Hexosamine Biosynthetic Pathway (HBP). Inhibition of PGM3 impairs N- and O-linked protein glycosylation, triggering activation of the Unfolded Protein Response (UPR) and intracellular ROS accumulation. Consequently, FR054 suppresses breast cancer cell proliferation, migration, and survival, while demonstrating potent antitumor activity in xenograft mouse models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
