Biological Activity

Fmoc-Val-Ala-PAB is a cathepsin B-cleavable dipeptide linker used in the construction of antibody-drug conjugates (ADCs) for targeted drug delivery. Upon receptor-mediated endocytosis and lysosomal trafficking, the valine-alanine sequence undergoes enzymatic cleavage by lysosomal cathepsin B, promoting intracellular release of the conjugated cytotoxic payload. This targeted cleavage mechanism enhances drug release within tumor cells while maintaining stability in circulation to induce targeted cytotoxicity and tumor regression. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
