Biological Activity

Roblitinib (FGF401) is a reversible covalent FGFR4 inhibitor with an IC50 ofand demonstrates over 1,000-fold selectivity across a panel of 65 kinases. By binding the FGFR4 kinase domain, it suppresses downstream FGF19-dependent signaling through the MAPK/ERK pathway. This target inhibition effectively impairs cell proliferation and induces cell death in FGFR4-activated hepatocellular carcinoma models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
