Biological Activity

Erlotinib (CP-358774) is a potent EGFR inhibitor (IC50 = 2 nM) that exhibits greater than 1000-fold selectivity for EGFR over human c-Src and v-Abl. By inhibiting EGFR tyrosine kinase activity and autophosphorylation, erlotinib blocks downstream cell survival signaling pathways. This inhibition suppresses tumor cell proliferation and induces cell cycle arrest and autophagy in sensitive cell lines. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
