EPZ015666 (GSK3235025) is a potent, selective, and orally bioavailable inhibitor of PRMT5 (Ki 5 nM), displaying over 20,000-fold selectivity over other protein methyltransferases. By blocking PRMT5 activity, it reduces symmetric dimethylation of histone and non-histone proteins, which leads to cell cycle arrest and suppression of cell proliferation in mantle cell lymphoma models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
