EPZ005687 is a potent and selective EZH2 inhibitor with a Ki of 24 nM in cell-free assays, demonstrating 50-fold selectivity over EZH1 and 500-fold selectivity against other protein methyltransferases. By inhibiting the enzymatic subunit of PRC2, EPZ005687 reduces global histone H3 lysine 27 trimethylation (H3K27me3) levels, leading to derepression of target genes and selective inhibition of cell proliferation and induction of apoptosis in EZH2-mutant lymphoma cells.
