Biological Activity

Enpatoran is a selective dual inhibitor of Toll-like receptor 7 (TLR7, IC50 = 11.1 nM) and Toll-like receptor 8 (TLR8, IC50 = 24.1 nM) in HEK293 cells. By blocking TLR7 and TLR8 activation by RNA-containing immune complexes, it suppresses downstream MyD88-dependent signaling and NF-kB activation. Consequently, it reduces the production of pro-inflammatory cytokines and type I interferons associated with autoimmune pathology.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
