Biological Activity

Enpatoran hydrochloride is a potent, orally active, dual Toll-like receptor 7 (TLR7) and Toll-like receptor 8 (TLR8) inhibitor. It targets single-stranded RNA sensing by suppressing TLR7/8 receptor activation, thereby blocking MyD88-mediated signaling pathways. Inhibition of this pathway impairs downstream NF-κB activation and interferon regulatory factor signaling, ultimately reducing the expression of proinflammatory cytokines and type I interferons involved in autoimmune inflammatory responses. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
