Enlicitide decanoate is an orally active macrocyclic peptide inhibitor of proprotein convertase subtilisin/kexin type 9 (PCSK9) with high binding affinity (Ki = 5 pM). By blocking the interaction between PCSK9 and the low-density lipoprotein receptor (LDLR), it prevents PCSK9-mediated LDLR degradation, thereby restoring hepatic cell-surface receptor density and enhancing the uptake and clearance of circulating LDL-cholesterol. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
