Endoxifen is an active metabolite of tamoxifen that acts as a potent estrogen receptor (ER) antagonist. In ER-positive MCF-7 breast cancer cells, endoxifen inhibits 17β-estradiol-induced expression of progesterone receptor (PR) mRNA. By binding to ERα, endoxifen suppresses ER-mediated transcriptional activation and downstream gene expression pathways, inducing receptor degradation and effectively inhibiting estrogen-dependent cell proliferation.
