Biological Activity
EN6 is a small-molecule covalent autophagy activator that directly targets cysteine 277 in the ATP6V1A subunit of the lysosomal V-ATPase with an IC50 offor recombinant human ATP6V1A. Covalent modification of ATP6V1A by EN6 decouples the V-ATPase complex from mTORC1, resulting in the inhibition of mTORC1 activity, enhanced lysosomal acidification, and activation of autophagic flux in mammalian cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
