Biological Activity
Emedastine is a potent, selective histamine receptor antagonist that targets H1-receptors (Ki 1.3 nM), exhibiting much lower affinity for H2-receptors (Ki 49,067 nM) and H3-receptors (Ki 12,430 nM). By blocking histamine binding, it inhibits histamine-stimulated intracellular calcium mobilization and downstream allergic inflammatory pathways. This antagonist activity effectively reduces histamine-induced vascular permeability, conjunctival hyperemia, and ocular inflammatory responses in preclinical models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
