Elacridar (GF120918) is a potent inhibitor of P-glycoprotein (P-gp/ABCB1) and breast cancer resistance protein (BCRP/ABCG2). By targeting these ATP-binding cassette efflux transporters, it blocks ATP-dependent drug transport across cellular membranes. This inhibition increases intracellular accumulation of chemotherapeutic substrates, effectively reversing multidrug resistance in resistant cancer cells and enhancing substrate bioavailability in preclinical models.
