Ordering Information

Biological Activity

Stenoparib (E7449) is an orally bioavailable, brain-penetrable small-molecule dual inhibitor targeting PARP1 (IC50 = 1.0 nM) and PARP2 (IC50 = 1.2 nM), while also inhibiting tankyrase 1 and 2 (TNKS1/2, PARP5a/5b). Through dual inhibition of PARP enzyme activity and tankyrase signaling, Stenoparib disrupts DNA repair mechanisms and modulates canonical Wnt/β-catenin pathway activity. These molecular actions lead to suppressed cellular proliferation and reduced tumor growth in DNA repair-deficient cancer models.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
