Biological Activity

E6446 is a potent inhibitor of Toll-like receptor 7 (TLR7) and Toll-like receptor 9 (TLR9) signaling. It directly interferes with nucleic acid binding by disrupting DNA-TLR9 interactions and blocking endosomal receptor activation. By suppressing downstream TLR7 and TLR9 pathways, E6446 attenuates NF-kB activation and proinflammatory cytokine production in various human and murine immune cell types, demonstrating significant immunomodulatory activity.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
