Asenapine is a multi-target receptor antagonist targeting serotonin receptors (pKi 8.4–10.5), adrenoceptors (pKi 8.9–9.5), dopamine receptors (pKi 8.9–9.4), and histamine receptors (pKi 8.2–9.0). High-affinity blockade of these G-protein–coupled receptors regulates central monoaminergic neurotransmission pathways, impacting downstream dopaminergic and serotonergic intracellular signaling cascades. In preclinical animal models, asenapine modulates neurotransmitter release and demonstrates potent antipsychotic and pro-cognitive behavioral phenotypes.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
