Biological Activity

Duloxetine (LY-248686) is a dual serotonin (5-HT) and norepinephrine (NE) reuptake inhibitor that preferentially inhibits 5-HT uptake over NE uptake in vitro and in vivo. By blocking serotonin and norepinephrine transporters, duloxetine elevates extracellular neurotransmitter concentrations in monoaminergic pathways. This inhibition modulates downstream monoaminergic neurotransmission and central pain pathways, producing antinociceptive and antidepressant-like behavioral phenotypes in animal models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
