DTNB (Ellman's reagent) is a non-fluorescent colorimetric probe used to quantify thiol group concentrations and acts as an allosteric inhibitor of dengue virus NS2B-NS3 protease and Streptomyces proteases. DTNB reacts with free sulfhydryl groups to form the yellow-colored product 2-nitro-5-thiobenzoic acid. Through covalent modification of accessible cysteine residues, DTNB disrupts protease-mediated enzymatic cleavage and viral polyprotein processing.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
