Biological Activity

DT-061 (SMAP) is an orally bioavailable small-molecule activator of protein phosphatase 2A (PP2A). By stabilizing specific PP2A heterotrimers, it promotes the dephosphorylation and degradation of key oncogenic drivers, including c-Myc and AKT. This activation impairs downstream AKT/mTOR and MAPK signaling cascades, thereby suppressing cell proliferation, decreasing cell viability in non-small cell lung cancer models, and inducing apoptosis in cancer cells. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
