Biological Activity

Taletrectinib (DS-6051b) is a potent, selective ROS1 and NTRK inhibitor targeting ROS1 (IC50 =, NTRK1 (IC50 =, NTRK2 (IC50 =, and NTRK3 (IC50 =. By suppressing ROS1 and TRK receptor tyrosine kinase phosphorylation, it blocks downstream MAPK/ERK and AKT/mTOR signaling pathways, resulting in the inhibition of cellular proliferation and the induction of apoptosis in ROS1- and NTRK-driven cancer models. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
