Biological Activity

DS-1001b is an orally bioavailable, brain-penetrating inhibitor that selectively targets mutant IDH1 R132X enzymes. By specifically inhibiting the mutant IDH1 enzyme, this compound blocks the abnormal conversion of alpha-ketoglutarate into the oncometabolite D-2-hydroxyglutarate. This metabolic blockade suppresses downstream oncogenic signaling, promotes cellular differentiation, and inhibits cell proliferation and tumor growth in mutant IDH1-driven cancers. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
