Doripenem is a carbapenem antibiotic with broad-spectrum antimicrobial activity against Gram-positive and Gram-negative bacteria. It inhibits bacterial cell wall synthesis by selectively binding to penicillin-binding proteins, particularly PBP 2 and PBP 3 in Pseudomonas aeruginosa and PBP 2 in Escherichia coli. This interaction disrupts peptidoglycan cross-linking during cell wall assembly, leading to bacterial cell wall degradation, morphological alterations, and bactericidal lysis.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
