T863 is a selective and orally active small-molecule inhibitor targeting acyl-CoA:diacylglycerol acyltransferase 1 (DGAT1). By interacting with the acyl-CoA binding site of DGAT1, T863 effectively blocks DGAT1-mediated triacylglycerol synthesis in cells. Inhibition of DGAT1 activity disrupts intracellular lipid accumulation and alters cellular lipid droplet formation, regulating key metabolic pathways involved in triacylglycerol synthesis and lipid storage. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
