DC260126 is a small organic antagonist targeting G-protein coupled receptor 40 (GPR40/FFAR1). It dose-dependently inhibits GPR40-mediated intracellular calcium elevations and ERK1/2 phosphorylation stimulated by free fatty acids, including linoleic acid and oleic acid. By disrupting GPR40-mediated pathway signaling, DC260126 suppresses fatty acid-potentiated glucose-stimulated insulin secretion and attenuates GPR40 mRNA expression in pancreatic beta cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
