Biological Activity
Dapansutrile (OLT1177) is an orally active, selective NLRP3 inflammasome inhibitor. By directly targeting the NLRP3 inflammasome, it inhibits NLRP3 oligomerization and prevents caspase-1 activation, blocking the processing and release of pro-inflammatory cytokines IL-1β and IL-18. This selective inhibition attenuates downstream inflammatory pathways in cell-based and animal disease models without impacting AIM2 or NLRC4 inflammasome activation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
