Biological Activity
CZC-54252 is a potent and selective small-molecule inhibitor of leucine-rich repeat kinase 2 (LRRK2), targeting both human wild-type LRRK2 and the pathogenic G2019S mutant variant. Pharmacological inhibition of LRRK2 kinase activity suppresses downstream cellular autophosphorylation and reduces phosphorylation of key Rab family GTPases, thereby regulating intracellular vesicular trafficking and neuronal autophagy signaling pathways. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
