Cycloleucine is a specific antagonist of the NMDA receptor coupled glycine receptor (Ki 600 μM). In addition, it acts as a specific inhibitor of S-adenosylmethionine-mediated methylation, blocking nucleic acid methyltransferase activity. By disrupting glycine site co-activation of NMDA receptors and impairing RNA methylation pathways, cycloleucine blocks essential cellular maturation mechanisms and exhibits potent cytostatic effects.[1][2]
