Biological Activity
CY-09 is a direct, selective NLRP3 inflammasome inhibitor that specifically binds to the ATP-binding motif of the NLRP3 NACHT domain. By blocking NLRP3 ATPase activity, CY-09 suppresses NLRP3 inflammasome oligomerization and assembly, thereby downstream preventing caspase-1 activation, interleukin-1β (IL-1β) cleavage, and pyroptosis. Publicly reported studies demonstrate that CY-09 exhibits potent anti-inflammatory effects in vivo, significantly attenuating disease severity in mouse models of gout, cryopyrin-associated periodic syndromes, and type 2 diabetes. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
