Biological Activity

CWI1-2 hydrochloride is a small-molecule inhibitor of insulin-like growth factor 2 mRNA-binding protein 2 (IGF2BP2). It binds IGF2BP2 and disrupts its binding to N6-methyladenosine (m6A)-modified RNA transcripts, thereby impairing glutamine metabolism pathways driven by target genes such as MYC, GPT2, and SLC1A5. This pathway disruption suppresses leukemia stem cell self-renewal and induces cell differentiation and apoptosis, demonstrating anti-leukemic activity in acute myeloid leukemia. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
