Culmerciclib (TQB3616) is a potent inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6) with highly effective antineoplastic activity. By suppressing CDK4/6 activity, it blocks Retinoblastoma (Rb) protein phosphorylation, arresting the cell cycle at the G1/S transition and inhibiting tumor cell proliferation. Culmerciclib demonstrates significant synergistic antitumor activity in estrogen receptor (ER)-positive and HER2-positive breast cancer models when combined with endocrine or HER2-targeted therapies. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
