Cucurbitacin D, a natural triterpenoid component of Cucurbita texana, functions as a small-molecule disruptor of the heat shock protein 90 (HSP90) chaperone machinery. It perturbs the HSP90 heteroprotein complex by disrupting interactions between HSP90 and its co-chaperones, Cdc37 and p23. Consequently, Cucurbitacin D inhibits HSP90 client protein maturation without triggering a pro-survival heat shock response, resulting in proteasomal degradation of downstream client proteins and suppression of cell proliferation. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
