CU-CPT22 is a Toll-like receptor 1/2 (TLR1/TLR2) complex inhibitor that selectively blocks Pam3CSK4-induced TLR1/2 activation without significant inhibition of TLR2/6. By preventing TLR1/2 heterodimerization, CU-CPT22 blocks downstream signaling cascades such as NF-kB activation, leading to decreased production of pro-inflammatory cytokines and attenuated immune responses. Tri-acylated lipopeptides like Pam3CSK4 mediate immune responses through TLR1/TLR2 heterodimer formation and signal activation. [1]
