Crenolanib (CP-868596) is a potent and selective receptor tyrosine kinase inhibitor targeting PDGFRα, PDGFRβ, and FLT3, including the imatinib-resistant PDGFRα D842V mutant. By blocking autophosphorylation of PDGFR and FLT3, crenolanib disrupts downstream signaling cascades, including the AKT/mTOR and MAPK/ERK pathways. This pathway inhibition leads to suppressed tumor cell proliferation, impaired angiogenesis, and induction of apoptosis in mutant-driven cancer models. [1][2]
