CQ31 is a small-molecule selective activator of the CARD8 inflammasome. It inhibits M24B aminopeptidases, specifically prolidase (PEPD) and Xaa-Pro aminopeptidase 1 (XPNPEP1), leading to the intracellular accumulation of proline-containing peptides. These accumulated peptides indirectly inhibit DPP8 and DPP9, triggering CARD8 autoproteolysis and assembly. Downstream activation of caspase-1 and cleavage of gasdermin D ultimately induce pyroptosis in targeted cells. [1]
