CPI-637 is a selective, cell-active benzodiazepinone bromodomain inhibitor of CBP (IC50 0.03 μM) and EP300 (IC50 0.051 μM). It exhibits high selectivity across the bromodomain family, demonstrating minor biochemical off-target activity only against BRD9. By blocking the interaction of CBP and EP300 with acetylated histones, CPI-637 suppresses chromatin reading and downstream transcriptional activation, leading to suppressed cellular proliferation and altered gene expression in cancer models.
