Biological Activity

CPI-169 is a potent and selective small-molecule inhibitor of histone methyltransferase EZH2 and its homolog EZH1. By blocking the catalytic subunit of Polycomb Repressive Complex 2 (PRC2), CPI-169 decreases tri-methylation of histone H3 at lysine 27 (H3K27me3), leading to the transcriptional activation of epigenetically silenced tumor suppressor genes and suppressed cancer cell proliferation. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
