Biological Activity
CPI-1612 is a potent, orally active inhibitor of EP300/CBP histone acetyltransferase (HAT). By inhibiting the catalytic HAT domain of EP300 and CBP, it blocks histone H3 lysine 27 (H3K27) acetylation and alters chromatin accessibility. This epigenetic blockade leads to the downregulation of downstream oncogenic transcriptional programs, suppressing cellular proliferation and exhibiting significant preclinical antitumor activity in mantle cell lymphoma and other malignancy models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
