Biological Activity

Lirametostat (CPI-1205) is an orally available selective inhibitor of EZH2 (IC50 = 2 nM) and EZH1 (IC50 = 52 nM). By targeting EZH2, the catalytic subunit of polycomb repressive complex 2 (PRC2), it selectively reduces cellular histone H3 lysine 27 trimethylation (H3K27me3) levels. This inhibition alters chromatin structure, derepresses silenced tumor suppressor genes, and impairs cancer cell proliferation and tumor growth in PRC2-dependent malignancies. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
