CP 43 is an ATP-competitive and selective inhibitor targeting TAO kinase 1 (TAOK1) and TAO kinase 2 (TAOK2). As a thousand-and-one amino acid kinase (TAOK) inhibitor, it blocks TAOK-mediated phosphorylation, thereby modulating downstream signaling within the p38 MAPK pathway. Inhibition of TAOK activity regulates microtubule dynamics, mitotic progression, and cellular stress responses, serving as a useful chemical tool for investigating TAO kinase-dependent cellular processes.
