Biological Activity

Coumarin 151 (7-Amino-4-(trifluoromethyl)coumarin) is a fluorogenic indicator and fluorescent tracer widely utilized for the detection and quantification of proteinase activities. Upon enzymatic cleavage of peptide substrates conjugated to this coumarin derivative, the released fluorophore exhibits strong fluorescence, enabling sensitive real-time monitoring of proteolytic cleavage in various biochemical assays. This fluorescent signaling strategy facilitates the characterization of protease kinetics, substrate specificity, and enzyme inhibition profiles across diverse enzymatic pathways.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
