Conduritol B epoxide is an irreversible covalent inhibitor of acid β-glucocerebrosidase (GCase). Pharmacological blockade of GCase disrupts lysosomal glycosphingolipid catabolism, resulting in cellular accumulation of glucosylceramide. This lysosomal impairment downstream increases neuronal α-synuclein accumulation. Consequently, conduritol B epoxide is widely utilized to model the neuropathological mechanisms of neuronopathic Gaucher disease and related synucleinopathies. [1][2]
