COH-SR4 is a small-molecule activator of AMP-activated protein kinase (AMPK) that suppresses adipogenesis in 3T3-L1 preadipocytes. Through indirect activation of AMPK, COH-SR4 modulates the mammalian target of rapamycin complex 1 (mTORC1) signaling pathway, which suppresses key regulators involved in protein synthesis and cell cycle progression. Downstream, this pathway inhibition downregulates transcription factors and target genes involved in lipid synthesis to prevent adipocyte differentiation and lipid accumulation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
