Coelenterazine h (2-Deoxycoelenterazine) is a synthetic luminophore derivative of coelenterazine that functions as a luminescent substrate for luciferases, such as Renilla luciferase (RLuc8), and photoproteins. Reconstitution of recombinant apoaequorin with coelenterazine h yields a photoprotein complex with enhanced luminescence output and increased calcium sensitivity. This photoprotein complex facilitates real-time monitoring of cytosolic calcium signaling and subtle intracellular calcium fluctuations in response to physical stimuli, such as touch and wounding. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
