Biological Activity

CM-272 is a first-in-class dual reversible inhibitor targeting G9a (IC50 = 8 nM), GLP (IC50 = 2 nM), DNMT1 (IC50 = 382 nM), DNMT3A (IC50 = 85 nM), and DNMT3B (IC50 = 1200 nM). Dual inhibition of G9a/GLP histone methyltransferases and DNA methyltransferases leads to epigenetic reprogramming, inducing immunogenic cell death and suppressing tumor progression to prolong survival in in vivo models of haematological malignancies.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
