CKI-7 is an ATP-competitive inhibitor of casein kinase 1 (CK1) that displays marked selectivity over other protein kinases, including CK2, PKC, CaMKII, and PKA. By targeted inhibition of CK1 activity, CKI-7 disrupts key signaling pathways such as Wnt/β-catenin and circadian clock regulation. This inhibition reduces downstream substrate phosphorylation, altering clock gene expression dynamics and suppressing cellular proliferation in various cell models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
