Cirtuvivint (SM08502) is an orally active, potent CDC-like kinase (CLK) and dual-specificity tyrosine-regulated kinase (DYRK) inhibitor that targets pre-mRNA splicing for Wnt pathway inhibition. By blocking CLK activity, cirtuvivint inhibits serine and arginine-rich splicing factor (SRSF) phosphorylation, disrupting spliceosome assembly and altering RNA splicing. Consequently, cirtuvivint downregulates Wnt pathway gene expression, represses Wnt signaling, and impairs tumor growth in solid tumor models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
