Cinobufagin is a natural cardiotonic steroid and active component of Venenum Bufonis that exhibits potent antitumor activity. It inhibits PI3K, AKT, and Bcl-2 signaling while triggering upregulation of ATM and Chk2, leading to downregulation of CDC25C, CDK1, and cyclin B. These molecular alterations disrupt cell cycle progression, inducing G2/M phase arrest and activating cleaved caspase-9 and caspase-3 to promote apoptotic cell death. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
