Biological Activity

CHZ868 is a type II JAK2 inhibitor that potently suppresses JAK2 signaling. By binding to the inactive conformation of the kinase, CHZ868 inhibits downstream STAT signaling pathways, including STAT1 and STAT5 phosphorylation. This targeted inhibition disrupts aberrant JAK/STAT signaling, resulting in suppressed cell proliferation and the induction of apoptosis in JAK2-mutated leukemic cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
