CHR-6494 is a potent Haspin kinase inhibitor with an ICvalue of 2 nM. By blocking Haspin activity, the compound prevents the phosphorylation of histone H3 at threonine 3 (H3T3ph) during mitosis. This loss of H3T3ph disrupts normal mitotic spindle assembly and chromosome alignment, triggering cell cycle arrest at the G2/M transition, inducing apoptosis, and suppressing cancer cell proliferation. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
